Application Area:

PCR with difficult DNA templates

We have finally found a perfect polymerase that works with the first PCR and does not need to be further optimized. We work a lot with plant DNA in the field of mangroves, macroalgae, seaweed. These templates in particular have many inhibitors (polyphenols and polysaccharides) and often make amplification difficult. NEB has a comparatively inexpensive alternative to the conventional, expensive polymerases.

Efficient PCR amplification for difficult templates.

PCR for recombinant DNA synthesis

The formulation that we purchased is the High Fidelity Hot Start Master Mix. Remarkably simple to use given the formulation without the need to pipette individual components. Accounting for the slightly higher annealing temperature needed for Q5, we were able to obtain several PCR products that were not amplified with our other favorite proofreading polymerase. It seemed to work particularly well with some of our genomic templates. Overall a great product.

With an error rate > 100-fold lower than that of Taq DNA Polymerase and 12-fold lower than that of Pyrococcus furiosus (Pfu) DNA Polymerase, Q5 High-Fidelity DNA Polymerase is ideal for cloning and can be used for long or difficult amplicons.

Q5 High-Fidelity DNA Polymerase is supplied with an optimized buffer system that allows robust amplification regardless of GC content. The 5X Q5 Reaction Buffer contains 2 mM MgCl2 at final (1X) reaction concentrations and is recommended for most routine applications.

For GC-rich targets (≥ 65% GC), amplification can be improved by the addition of the 5X Q5 High GC Enhancer. Q5 High-Fidelity DNA Polymerase is unlike typical, lower fidelity PCR enzymes.
