Prometheus Panta + RA for High Throughput Stability Characterization

NanoTemper TechnologiesAvailable: Worldwide

Prometheus Panta + RA monitors the thermal stability, turbidity, and dispersity of a protein sample. It also measures the hydrodynamic radius, molecular weight, and self-association propensity of your protein-based therapeutics for developability profiling. The Panta uses dye-free, buffer-independent measurements from nanoDSF (nano Differential Scanning Fluorimetry), DLS (Dynamic Light Scattering), and backreflection technolo…

Use only 10 µL of sample to obtain high-quality data that enables you to make better decisions about your buffer conditions and biologic candidates.

The Prometheus Panta measures thermal unfolding and turbidity in parallel using near-UV detection systems. The high-quality data from nanoDSF, DLS, and backreflection technologies is gathered in parallel, along an entire thermal gradient, enabling you to make better decisions about the stability of your protein formulations while using small volumes of sample. Great for researchers doing HTP screening of small molecule thermal-shift assays, buffer formulations, developability profiling, or mutational analysis.

The RA (robotic autosampler) enables you to prepare four 384-well plates (1536 samples), which are loaded up and automatically measured by the system, hands-free. With the NT.RA, improve your throughput without sacrificing data quality.

Key Features:

  • Label-free measurements in solution
  • Broad dynamic range, as low as 0.02 mg/ml to >200 mg/ml protein
  • Measure thermal or chemical stability using intrinsic fluorescence of your proteins with nanoDSF dual UV system: 330 nm and 350 nm fluorescence detection
  • Measure particle size and dispersity with visible light DLS
  • Monitor amorphous protein aggregates with backreflection, or turbidity detection
  • Capillaries hold 10 µl of sample; 24 capillaries mounted on a chip are automatically loaded with the robotic arm, enabling you to walk away while up to 1536 data points are collected
  • nanoDSF parameters: Tm, Tonset, reversibility of unfolding
  • DLS/SLS parameters: rH (hydrodynamic radius), PDI (polydisperisty index), B22, kD, Tsize
  • Backreflection parameters: Tturbidity (onset of turbidity, particles >12.5nm in radius)
  • Temperature range: from 15 °C to 110 °C
  • Straightforward handling: simple sample preparation and intuitive software user interface for both small-scale and high-throughput experiments
