Description

The vast amount of host DNA compared to microbial DNA in clinical samples often reduces the sensitivity in molecular analyses of microorganisms. MolYsis™ Basic5 kit is used prior to DNA isolation to deplete the host DNA and enrich the intact pathogen cells from small and large sample volumes. This leads to increased analytical sensitivity and can be combined with any commercial DNA extraction and isolation kits, manual or automated. With MolYsis™ Basic5 human or animal cells are selectively lysed and free DNA is degraded in a DNase treatment step. Afterwards a reagent, BugLysis, is used for the broad-range lyses of Gram-positive and Gram-negative bacteria and fungi. The enriched microbial DNA can be isolated and purified with the method of choice. Applications comprise next generation sequencing, amplicon cloning, real-time PCR, Sanger sequencing and DNA array analysis of microorganisms.

Depletion of host DNA significantly improved microbial analysis

In this product brochure, Molzym introduces MolYsis, which tackles the issue of limited microbial DNA presence in complex human or animal samples compared to host DNA. MolYsis employs a proprietary technology to selectively lyse host cells, leading to up to 99% reduction in host DNA. A subsequent lysis step targets intact bacteria and fungi cells, resulting in purified microbial DNA. This process can be conducted manually or automated using the SelectNA™ plus device. The enhanced microbe-to-host DNA ratio achieved through this method significantly enhances sensitivity in subsequent NGS, PCR, and other molecular microbiological techniques.

Selective Enrichment of Bacterial and Fungal DNA

MolYsis™ is a tool for the removal of human DNA and enrichment of bacteria and fungi from liquid samples. Technically, in a short series of steps, MolYsis™ effects the selective lysis of blood cells and the quantitative degradation (>99%) of released human DNA. After enrichment from the lysate by centrifugation, microorganisms are treated with a reagent degrading cell walls of Grampositive and Gram-negative bacteria and fungi. The broad range lysis potential has been shown in clinical evaluations, including over 200 genera of bacteria (86 Grampositives, 120 Gram-negatives) and 65 genera of fungi.
