Reporter gene assays: Transcription factor activity

The Centro LB 960 was acquired back in 2004 for our lab and was in use for several years. At some time people stopped using it and it was stashed in the storage. By now, 10 years without a purpose our luminometer is needed for some luciferase based reporter assays and we wondered if the Centro is still fit for service. With only a little effort our sales representative could install drivers and software on a new PC (Win7) and confirm that our Centro worked flawlessly. We did not expect it to be operational after such a long period of time. The hardware is obviously very robust and working with the software interface is intuitive. We can't wait to use our Centro for producing good data.

The Centro LB 960 is easy to program and user-friendly. It automatically generates a spreadsheet that can be exported to Excel. It has a small footprint so doesn't take up much bench space. Their customer service is excellent - you can always email a technical expert for help and their representative is very helpful.

The Centro is a high-performance, easy to use microplate luminometer for both, flash and glow luminescence applications. The optimized optical system provides lowest background and negligible crosstalk (10-6). Detection of parasitic molecular strategies to influence host plant signaling

Parasitic plants of the genus Cuscuta are obligate holoparasites with a broad host spectrum for nearly all dicotyledonous plants. As leafless and rootless plants, Cuscuta spp. wind around stems of host plants and penetrate host tissue with haustoria. They directly connect to the vasculature and withdraw water, nutrients, and carbohydrates. Thus, the haustorium development and the establishment of a connection to the host represent essential steps in the parasite’s life cycle.

Molecular cues might be necessary to influence intracellular processes and the connection to the host vascular system. To identify such molecular cues, promotors of host genes that are upregulated at the Cuscuta infection site can be fused to a luciferase reporter gene.
