Separation of secondary metabolites, protocol development
The system fails at higher pressure. Always problems with synchronization of sample loop with injection. Fraction collector is good but resolution of peaks are not perfect to ensure separation. Communication issues with RS232 and USB for the auxiliary pump.

A preparative HPLC system should be as versatile as possible, since purification tasks can change frequently. AZURA Preparative HPLC was designed for flexibility and to comfortably handle large sample volumes. Extensive automation makes fraction collection easy and allows for solvent as well as peak recycling.
AZURA Preparative HPLC features a pulsation compensated pump drive with selectable constant flow or constant pressure solvent delivery. Supported flow rates range from 0.1 to 1000 ml/min. Automated piston seal wash helps extending seal life, especially when working with buffers. Pump heads are available in different materials and capacities, all featuring RFID recognition, which is not just comfortable, but also GLP compliant and it helps meeting maintenance intervals. A sophisticated leak management and detection is built-in.
How do you stay up-to-date?
AZURA Preparative HPLC systems can be supplemented with many functional options starting with online degassing and continuing with gradient support, large volume injection, solvent switching, column switching, detectors, including remote flow cell versions, cascaded fractionation, and more.
Flexibility extends to the control options: normally run by software, the tablet controlled Mobile Control is also available for autonomous operation. Within the software direct control is always possible, even while running a method.

In this application note, KNAUER describes a gradient method designed to provide fast determination of six steviol glycosides using reversed-phase HPLC and UV detection. An automated matrix reduction is achieved by online SPE (solid phase extraction), aiming to speed up sample preparation and promise a high sample throughput.
Scale-Up of an Analytical HPLC Method for Steviol Glycosides to a Preparative Approach
In this application note, KNAUER describes how an existing analytical HPLC method can be transformed to a preparative HPLC using the KNAUER Scale-Up Converter.
