Efficient, reproducible, and scalable NGS library preparation is essential for producing high-quality sequencing data. But as NGS workflows become more complex — incorporating diverse sample types, low-input libraries, and higher-throughput demands — the need for automation has never been greater. Manual pipetting introduces variability, increases error risk, and limits scalability.

Revvity’s suite of automated liquid handling NGS workstations — including the Fontus™, Sciclone™, Zephyr™, and BioQule™ systems — provides a comprehensive range of throughput options to suit diverse NGS needs. These platforms, combined with over 120 vendor-qualified automated methods, empower researchers to move from installation to sequencing-ready samples in days, not months.

Automating liquid handling in NGS library prep brings a range of benefits: Increased throughput reduces library prep bottlenecks, boosting productivity in your NGS projects. Extended walkaway time allows researchers to focus on other critical tasks, which is especially valuable for time-intensive NGS workflows. Meanwhile, automation also significantly boosts reproducibility, ensuring consistent results and enhancing confidence in sequencing data.

Revvity's automated NGS library preparation workstations by throughput

Revvity's range of automated NGS library preparation workstations, ordered by throughput capabilities

Fontus NGS Workstation

The Fontus NGS workstation delivers the highest level of throughput and walkaway automation in Revvity's NGS workstation portfolio. Capable of processing up to 384 samples simultaneously, it offers full flexibility for high-throughput NGS workflows. The Varispan™ pipetting arm allows for tube-to-plate reformatting, fast normalization, and sample pooling, all while reducing reagent costs and hands-on time. This system is ideal for large-scale sequencing projects that require greater efficiency.

Deck Verification is an optional vision system for Fontus workstations and acts as an intelligent safeguard to reduce desk setup errors. Deck Verification indicates if labware is correctly positioned before and during a run. Using an overhead camera and recognition software, it detects misplaced consumables, incorrect labware and missing accessories, preventing workflow disruption and costly reruns.

Sciclone G3 NGSx and NGSx iQ Workstations

The Sciclone G3 NGSx is a high-precision system tailored for large NGS workflows. The NGSx iQ version integrates an on-deck thermal cycler (ODTC) and the Twister™ III Robotic Arm, enabling complete PCR and incubation automation, eliminating off-deck handling. This solution is perfect for high-throughput workflows where walkaway time is paramount.

Zephyr G3 NGS Workstation

For labs seeking a compact yet powerful option, the Zephyr G3 workstation offers a smaller footprint while supporting up to 96 samples per run. This system features a 96-channel pipetting head, on-deck temperature control, and integrated shaker, making it an agile choice for medium-throughput NGS operations.

Powering workflows with vendor-qualified methods

One of the most significant hurdles in NGS automation is the time and effort required to develop and validate custom methods for each library preparation kit. Revvity addresses this problem with over 120 vendor-qualified automated methods, pre-validated and ready to deploy. These methods offer a plug-and-play experience, enabling labs to fast track from installation to sequencing in as little as five days.

Drastically reducing the need for manual programming or custom protocol development, these methods are pre-built, QC-tested, and vendor-approved to meet kit performance standards. Revvity's automated methods undergo a rigorous verification process, including water and chemistry testing, to ensure compatibility with best-in-class NGS kits. Each method is optimized to match or exceed vendor benchmarks, delivering highly reproducible, high-quality sequencing results you can rely on.

Some of the vendor-qualified methods include library prep kits from:

  • Illumina®
  • New England Biolabs (NEB®)
  • Integrated DNA Technologies, Inc. (IDT®)
  • Pacific Biosciences (PacBio®)
  • QIAGEN®
  • Twist® Biosciences
  • Watchmaker® Genomics

These are just a few of Revvity's qualified kit vendors. Explore the full list here.

How do vendor-qualified methods benefit NGS workflows?

  • Rapid start-up: Move from system installation to sequencing-ready libraries in as little as 5 days, reducing start-up times from months to days.
  • High-quality results: Vendor-validated methods ensure compliance with kit specifications, resulting in high-quality sequencing data with minimal optimization.
  • Reduced variability: With pre-tested protocols, each sample receives uniform handling, lowering the risk of batch-to-batch variation in sequencing results.
  • Continuous compatibility: As kit manufacturers release new products, Revvity continuously updates its automated methods, ensuring seamless compatibility with evolving NGS kits.
