Huntington's disease (HD), a neurodegenerative disorder characterized by loss of striatal neurons, is caused by an expansion of a polyglutamine tract in the HD protein huntingtin. This gene encodes a protein belonging to a class of huntingtin interacting proteins characterized by WW motifs. This protein is a histone methyltransferase that is specific for lysine-36 of histone H3, and methylation of this residue is associated with active chromatin. This protein also contains a novel transcriptional activation domain and has been found associated with hyperphosphorylated RNA polymerase II.
Good antibodies for detection of SETD2 by western blot!
Western blot
I used these antibodies with cell lysates extracted from lung cancer cell lines (A549 and H1299). The antibodies give good signal at about 300 kDa, but have a little bit of non-specific binding at about 100 kDa. For detection of SETD2 with these antibodies I used regular western blot protocol for high molecular weight proteins (8% PAAG and wet transfer overnight). Antibodies work well for overnight incubation +4C, and room temperature incubation for 1.5-2 h. I can recommend these antibodies! Few companies make antibodies for this protein.
