Strong intensity, low background, great value
Immunofluorescence
I use this product to verify that human primary macrophages reached maturity (derived from peripheral blood of healthy donors). Staining is always consistent, strong and specific. The protocol we use is briefly as follows: Human primary macrophages are encapsulated in 3D hydrogel composed of hyaluronic acid and collagen type 1. After 5 days in culture, the hydrogels are fixed with 4%PFA for 10 minutes, permeabilized with 3 x 5 min washes of TBSTriton X 0.25%. Then, blocked with TBS 10% goat serum, 1% BSA for 1 hour at RT. Then primary antibody anti-CD68 (333802) is used at 1:100, incubated overnight at 4*C. After washes, secondary antibody goat anti-mouse AlexaFluor 488 is used at 1:500 for 1 hour at RT. Images are acquired using a Nikon A1 Confocal Microscope.
