Tricky but specific.
Western blot
"Dirty antibody", do not use BSA for the blot. Use only 5% milk in TBST. Use TBS, Tween 0.2% instead of 0.1% to get cleaner membranes. dilutions: 1/1000 for primary, 1/5000 for secondary. Antibody is however specific. In this specific tissue, it shows 3 bands, the main band at 100kDa "full lenght", two light bands at 70kDa and 26kDa "N-ter cleaved" and C-ter cleaved respectively (theorically). These tree bands seem specific, cause my KO tissues show a significant increase in PCDHG expression (RNA-Seq data). Indeed, WB using this antibody confirmed their increased protein level. all 3 bands were higher in my KO tissues compared to WTs.
