When ligating DNA fragments over 10 kb in length, the ligation efficiency will be very poor if a conventional ligation kit is used. DNA Ligation Kit LONG includes an optimized ligase/buffer system that enables you to ligate long DNA fragments without need for specialized expertise or extensive optimization. This application note describes the use of TaKaRa DNA Ligation Kit LONG to construct a BAC library with genomic DNA inserts from mammalian blood samples.
