Screening for Use-Dependent Blockers of NaV1.5 Channels on the IonWorks Barracuda Automated Patch Clamp System
18 Nov 2013This application note describes the design and validation of a cell based assay for the NaV1.5 channel, using the IonWorks Barracuda® Automated Patch Clamp System. A single concentration mock screen of known sodium channel blockers was set up to identify the location of the hits in a 384 compound plate. Follow-up experiments on the hits were then performed to identify use- and state-dependent blockade using a train of depolarizing pulses.

ElectrophysiologyElectrophysiology is the study of biological voltage changes, from a single ion channel through to a whole organ scale. Voltages can be measured using patch clamping, automated workstations, electrophysiology rigs or cell based assays. Specific cell lines, manipulators and microinjectors are also available for electrophysiology research.Ion Channel
