Multiplex Measurement of Rat Kidney Cytokines in Serum and Urine using the Fluorokine® MAP

6 Aug 2012

This application note investigates kidney toxicity protein markers by measuring TIM-1/KIM-1/HAVCR, FABP1/L-FABP, Lipocalin-2/NGAL, Cystatin C, and Osteopontin in serum and urine from normal (Sprague Dawley®) and PCK rats using the Fluorokine® MAP Rat Kidney Toxicity Panel.

Fluorokine MAP Kits for the Luminex platform are highly efficient multiplex assays that have been validated for the quantification of factors in complex matrices and require little or no optimization. For a wider selection of analytes, please see our VersaMAP™ line of development kits validated for use with cell culture supernatants. Features Simultaneously detect multiple analytes Requires small sample volume (Undergoes similar validation testing as the Quantikine® single analyte assays Complete, ready-to-use kits Available in three formats. For details, see Which Bead-based Multiplex Kit is right for me? 1. Custom Premix Kits • Custom ordering tool is used to select the analytes of interest and to submit your order • User defines up to 10 analytes of interest • Bead and Detection Antibody cocktails are sent premixed in respective vials • Quick turn-around time: only 2 - 4 days Please allow additional time when placing orders through a distributor. 2. Custom Base Kits & Bead Sets • Base Kit and Bead Sets are provided separately and mixed by the end-user • Customized for the detection up to 22 analytes, depending on the Base Kit selected • Rapid delivery: typically next day Please allow additional time when placing orders through a distributor. 3. Predefined Kits • Panels are predefined by R&D Systems • Complete, ready-to-use kits • Bead and Detection Antibody cocktails are provided premixed in respective vials • Rapid Delivery: Typically available next day Please allow additional time when placing orders through a distributor. ELISAEnzyme-linked immunosorbent assays (ELISA), also known as enzyme immunoassays (EIA), are used for the detection and quantification of proteins, peptides and antibodies in a sample. ELISA often comes in kit format, with pre-selected antibody pairs to detect specific proteins or biomarkers of diseases such as diabetes and obesity, or cardiovascular and neurological disorders. Detection is made possible with chemiluminescent, fluorescent or colorimetric substrates and detection instruments such as microplate / ELISA readers. Other kits available include: Enzyme-linked immunospot (ELISPOT) assays for the quantification of protein-producing cells and chemiluminescent immunoassays (CLIA) kits. Find bioassays for absorption, distribution, metabolism, and excretion of drug molecules including cytotoxicity, transporter/permeability, metabolism and activity assays as well as hepatocytes and cell lines for ADME. This can reveal information on cell health (apoptosis, cytotoxicity, viability and proliferation assays), cell metabolism, cell migration and cell signaling mechanisms. The use of biomarkers in research and diagnosis can indicate a normal or disease state or drug response of cells / tissues. Biomarkers include genetic markers, cell surface markers such as antigens, antibodies or receptors and secreted molecules such as cytokines. An assay system is required for identification of biomarkers. :Clinical ChemistryBiochemistry (or clinical chemistry) involves the analysis of bodily fluids using chemical tests. Techniques used include HPLC, chromatography, spectroscopy, mass spectrometry, immunochemical, electrophoresis, turbidometric / spectrophotometric assay, MRI and ISE analysis. Tests are often carried out on plasma or serum but urine (urinalysis) and fecal specimens are also processed.Toxicology / Drugs of Abuse TestingToxicology and Drugs of Abuse Testing is the investigation into toxic and illegal substances found at the scene of a crime or from a suspect and / or victim associated with a scene of a crime. Analyzers, immunoassays and drugs of abuse test kits and presumptive test kits are an essential aspect of toxicology investigation.Kidney Damage
