Monitoring Enzyme Catalysis using the VERTEX 80 FT-IR Spectrometer in Rapid Scan Mode

25 Oct 2011

This application note describes the monitoring of the peptide product of the chymotrypsin-catalysed hydrolysis of N-succinyl-Ala-Ala-Pro-Phe-p-nitroanilide (Suc-AAPF-pNA) by FT-IR spectroscopy. This reaction can be followed by FT-IR due to the formation of a new C-terminal carboxylate group. The VERTEX 80 FT-IR spectrometer was shown to collect more than 100 spectra at resolution 16 cm-1 per second at the fastest mirror velocity (320 kHz).

Infrared / IR SpectroscopyInfrared (IR) spectroscopy measures the interaction of infrared light with a sample, including transmission, reflectance & absorbance, facilitating the identification of analytes. Equipment used for quantitative analysis includes Fourier-transform infrared (FTIR) spectrometers, infrared cameras, FTIR gas analyzers, as well as attenuated total reflectance (ATR) accessories and pellet or film presses. Measurements can be conducted using a Fourier-transform near infrared (FT-NIR) spectrometer, while there are also UV-Vis- NIR spectrophotometers that measure a broader spectrum of wavelengths.
