Genomic DNA Isolation from Whole Blood using AcroPrep™ Advance 96-Well DNA Binding Plates on an Automated Liquid-Handling Robot

12 Apr 2012

The protocol in this note represents an example of a fully automated process for high throughput sample preparation from human blood (anticoagulated) optimized for DNA binding plates, and using commercially available buffers with a liquid handling system.

AcroPrep™ Advance Filter Plates

Pall Life Sciences - Laboratory, Food, Beverage

AcroPrep™ Advance filter plates are the newest edition to Pall’s filter plate family. New sealing technology and advanced tip design make these the first choice in critical sample prep applications.   Aplications: Lysate Clearance DNA purification Protein purification Multiplex assays Aqueous filtration Particulate removal Size exclusion Bead-based assays Neonatal screening Flow cytometry Solvent filtration Concentration, purification and desalting of proteins and nucleic acids    Features: Smooth well design – Provides consistency in filtration times, as well as efficient sample and bead recovery Optimized outlet tips – Minimizes sample leakage during incubation steps and reduces the presence of hanging drops following filtration New well geometry – Results in faster, more uniform filtration rates across the plate, thereby improving well-to-well, plate-to-plate and lot-to-lot consistency 100% visual inspection – Ensures each well has proper membrane placement and is free from debris Automation compatible – Manufactured in accordance with SBS guidelines, allowing plates to be run in manual, semi-automated and automated processes

Genome AnalysisGenomics, the study of genomes, includes functional genomics, evolutionary genomics and comparative genomics. There are many genomic technologies such as DNA sequencing of whole genomes, computational biology and bioinformatics. DNA and nucleic acids must be isolated and concentrated from cells for analysis with kits, automated analyzers and software. Other useful technologies for studying genomics include PCR, microarrays and electrophoresis.DNA / RNA Extraction and PurificationPurified DNA and RNA are required for numerous downstream molecular biology applications. Consequently, the importance of high-quality DNA/RNA extraction and purification equipment cannot be underestimated. Many purification kits are available and are typically optimized for nucleic acid type and source, including plasmid DNA, genomic DNA, mRNA, RNA and viral nucleic acid purification kits. Automated extraction and purification of nucleic acids can be implemented with magnetic bead separator instruments or high-throughput purification workstations.
