A Fully Automated, Bottom-up Approach for MALDI-TOF MS Based Discovery Workflows

This application note demonstrates a fully automated solution for MALDI-TOF MS based peptide mapping experiments. Using an online digestion, desalt, reversed phase separation and fraction collection platform, the average experimental time was decreased from over 18 hours to less than 30 min (reversed phase separation included – acquisition time not included) while substantially improving sequence coverage.

AccuSpot

The AccuSpot automates LC microfractionaction, spotting and preparation for MS analysis. With the AccuSpot system, LC eluent can be accurately and continuously spotted onto target plates, in trace amounts of 50nL or more. This allows for automatic preparation of target plated for MALDI-TOF-MS measurements. By using it in conjunction with Shimadzu's 1D or 2D Micro LC System and Shimadzu Biotech's AXIMA MALDI-TOF instruments, proteome analysis can be performed with a higher degree of precision and with greater sequence coverage.
