Description

The NightSHADE evo LB 985N In Vivo Plant Imaging System is a modular, easy to use optical imaging system dedicated to in vivo analysis of plants. Equipped with an absolutely lighttight cabinet and a deeply cooled CCD camera it enables sensitive luminescence and fluorescence monitoring in tissues, seedlings and whole plants.

The camera can be attached either to the top or the side of the darkroom – the sample chamber – to enable imaging from above and from the side. Furthermore, key environmental conditions like temperature or light can be simulated to provide a controlled growth environment.

The usage of fluorophores such as YFP and GFP is a widely distributed and established method in plant research. To monitor fluorescent reporter proteins several techniques based on instruments such as fluorescent microscopes or imaging systems are available. Detection of free or fused fluorescent reporters in different subcellular localizations and with different expression levels can be performed with the NightSHADE LB985 imaging system. The obtained results, presented in this application note, correlated with data received by detection of fluorescent using the Mithras LB940 multimode plate reader.

Analysis of the Circadian Clock and Plant Growth with NightSHADE

In this application note by BERTHOLD TECHNOLOGIES, gene expression in Arabidopsis is analysed using promoter::firefly Luciferase (LUC) reporter constructs and the NightSHADE imaging chamber. Analysis of circadian clock associated 1 (CCA1) and chlorophyll a/b-binding protein (CAB) constructs showed that CCA1 expression preceded activation of CAB promoter, consistent with CCA1 protein playing a positive role in CAB gene expression.
