Cryolys Evolution - Cooling System for Thermo-Sensitive Samples
Cryolys Evolution is a patented cooling system compatible with Precellys Evolution tissue homogenizer. It prevents thermo-sensitive samples from heat degradation during homogenization process.
Serves its purpose very well, but could use some improvements
RNA isolation for gene expression analysis by qPCR
The functions of the Precellys evolution are great for our purposes, which mostly involve homogenization of various tissues for the purposes in Trizol for subsequent RNA isolation and gene expression analysis. However, I have a few grievances. As many scientists use Trizol/Tri-reagent for the purposes mentioned above, I was very disappointed to find that the lid and sample racks of the Precellys are not Trizol-proof, meaning they've developed many pockmarks and streaks because no matter how well you have tightened the homogenization tubes, there is an occasional leak. Additionally, the 7mL and 15mL tubes that the Precellys uses are great for their instrument, however, there are very few commercially available racks to stably hold large numbers of tubes (>20). There are centrifuge adaptors for centrifugation of the 7mL tubes, however there is not a corresponding adaptor for the 15mL tubes. The 15mL tubes fit into a 50mL conical adaptor for a bucket rotor, however, getting them back out is a problem. The "solution" of dumping the Precellys tubes from the bucket rotor is counter-productive to the centrifugation just performed.

Best practices in protein sample preparation with Precellys Evolution
For thermo-sensitive molecules such as proteins, sample preparation is a critical step before proceeding to molecular downstream analysis. In this application note, Bertin Instruments highlights the key considerations in protein sample preparation.
Precipitation of Proteins and Lipid Extraction From Homogenized Rat Skin Tissue
In this study, protein precipitation and bioactive lipid extraction from rat skin tissues is reported. The combination Solid Phase Extraction and liquid chromatography-tandem mass spectrometry (LCMS/MS) approach was used to measure concentrations of lipid mediators in these tissues. Rat skin tissues were homogenized using CKMix50_7mL Precellys® Lysing Kit and Precellys® Evolution tissue homogenizer combined with Cryolys® cooling unit. In addition, temperatures of homogenates were measured to investigate if temperature of complete homogenate of rat skin tissues remained below 40°C.
Best Practices for Tough Sample Grinding in Molecular Biology Using Precellys
The biggest challenge when working with fibrous, elastic and tough samples is efficient sample processing. Furthermore, generation of heat causes degradation and denaturation. Here, Bertin Instruments describes the improvement of extraction yields of tough samples using the Precellys® evolution homogenizer, thanks to 3D-bead beating technology. When paired with the cryolys evolution cooling unit, this technology can also prevent degradation of heat-sensitive biological samples.
