Purification of Adeno-Associated Virus using Affinity Chromatography with Sartobind® Membrane Adsorbers
Adeno-associated viruses (AAV) belong to the family Parvoviridae, subfamily Parvovirus, genus Dependovirus. They are used as gene delivery vectors for human gene therapy. The advantages of AAV as a vector include e.g. its lack of pathogenicity, high titer, ease of manipulation, and ability to infect non-dividing cells. This application note describes the purification of adeno-associated virus using affinity chromatography with Sartobind® Membrane Adsorbers.
Separation of Proteins and Peptides with Sartobind® Membrane Adsorbers
In this application note, ion exchange chromatography and Sartobind® membrane adsorbers were used to separate protein and peptide mixtures. All samples were prefiltered with a Minisart filter having 0.2 μm or a 0.45 μm pore size.
Isolation of DNA Fragments from Agarose Gels Using Sartobind® D Anion Exchanger
The isolation of DNA fragments from agarose gels is a standard method frequently used in molecular biology. A variety of isolation methods have been developed, all with advantages and disadvantages. This procedure describes the use of Sartobind D membranes for electrophoretic isolation of DNA fragments by insertion of small pieces of the weak anion exchange membrane into the agarose gel. It has the advantage of being inexpensive, efficient and has no contamination of agarose in the purified fragment.
