A simple machine to automate route PCR pipetting. Software needs to be improved a bit
PCR / Real time set up
We got a Pipetmax 268 system as an automatic liquid handling system to automate our PCR pipetting for conventional and real time PCR setup in 96 well plate format. The system is compact, does not occupy too much bench space and is controlled by a Pipetmax Software installed in a compact touchscreen laptop. We were able to automate key PCR plate pipeting for qPCR from 1.5 ml tubes to 96 well plates., 1.5 ml tubes to 8 well strips etc. The software is intuitive, guides the user visually to place the various consumables ( Tips, Tubes , PCR plates, Waste bin) etc at the respective positions. Also there is an option to input the number of missing tips from left or right in an already used tip rack. The software also has a simulation mode which simulates the pipetting procedure in the Pipetmax run before the actual run. This is useful for testing new protocols before placing the actual samples on the Pipetmax. Upon testing the machine for qPCR we found the Ct values to be more consistent with Pipetmax, when compared to manual pipetting. The Pipetmax accepts most standard Gilson pipet tips apart from the racked tip in blister pouches for pipetmax. Most of our pipetting protocols were sent to the Gilson specialist and they converted the protocols into Pipetmax protocols incorporating the specifications of the consumables at our end. These were uploaded into the machine via the USB provided. As an end user, we found this limiting. We would like the software to allow the user to make custom protocols utilising the different consumable specifications listed in the software library. Gilson lists a Protocol Buildler software and life time license in the Related Products at the website. I feel this software also should be provided free of cost to all users of Gilson Pipetmax so that end users can custom build/ modify their own protocols rather that approach Gislon customer support each time for the same purpose.

Automation of the Agencourt AMPure XP PCR purification system
In this application note, Gilson introduces an automated method for the Agencourt® AMPure® XP PCR purification system using PIPETMAX®. This method simplifies and enhances the reliability and flexibility of a typically laborious and technically challenging process. Genomic DNA from Escherichia coli was prepared for whole genome sequencing using the Illumina® Nextera® XT DNA Library Preparation Kit. The Agencourt AMPure XP PCR purification system is employed to clean up the library post-amplification and before normalization. A custom magnetic bead separator rack allows efficient purification of up to 96 samples in a single run. The automated protocol is adjustable, accommodating various factors like sample quantity, volume, bead volume, wash steps, and incubation times to suit specific application needs. This application enables seamless integration of PIPETMAX into diverse workflows.
