When selecting a buffer for your NMR sample, it is best to use a deuterated buffer instead of one that's protonated. Doing so reduces the contribution of proton signals from the buffer in the 1H-NMR spectrum from the sample containing the buffer without affecting the buffering capacity. Likewise, the best way to reduce proton signals in 1H-NMR from the use of common protonated reagents is to use deuterated reagents, such as deuterated acids, bases, and reducing agents. Additionally, utilizing deuterated detergents and phospholipids in preparing membrane protein samples not only helps ensure the protein assumes the correct three-dimensional fold and remains active, but also minimizes unwanted signals from appearing in the spectrum.
