![Western Blot: c Abl [p Tyr412] Antibody [NSB252] - Extracts of fibroblasts infected w/ oncogenic dSH3-Abl (1-4) or recombinant phosphorylated c-Abl (5, 6) were resolved by SDS-PAGE on a 10% Tris-glycine gel and transferred to PVDF. The membrane was either treated with YOP phosphatase (5) or untreated (1-4, 6), blocked with 5% BSA-TBST buffer ON at 4C, and then incubated with Ab for 2hrs at RT in a 3% BSA-TBST buffer, following prior incubation w/ no peptide (1, 5, 6), non-phosphopeptide corresponding to the immunogen (2), generic phosphotyrosine containing peptide (3), the phosphopeptide immunogen (4). Only the peptide corresponding to NSB252 blocks the Ab signal, demonstrating the specificity of the Ab.
