Long-Term Hepatotoxicity Studies Using Cultured Human iPSC-Derived Hepatocytes

Drug-induced liver injury (DILI) continues to be a growing public health problem and a challenge for drug development. In vivo models remain the gold standard to test for potential DILI. However, these studies are costly, time-consuming, and more importantly, rather poor predictors of human toxicity. 2D hepatocyte cultures can be used as an alternative to animal models, yet many limitations such as high variability, changes in cell morphology as well as liver-specific functions during long-term culture, remain an issue for in vitro models. In this application note, we demonstrate the suitability of 3D cultured human iPSC-derived hepatocytes for hepatotoxicity studies, which aims to deliver a highly accurate, long-term in vitro model for DILI tests.
