A Robust and Sensitive Capillary Flow LC – High-Resolution Accurate-Mass MS Platform for Discovery and Targeted Proteomics

23 Oct 2016

This poster describes a reliable and easy-to-setup capLC-MS platform for sensitive targeted and shotgun analysis of peptides and proteins. The obtained results were compared with nano and analytical flow LC-MS analysis and recommendations for optimal LC and ESI interface configuration for different proteomics applications were formulated. The optimized capLC-MS conditions resulted in high-throughput separations with ~ 10 min total analysis time for targeted quantification of peptides at amole level.

Thermo Scientific™ Q Exactive™ BioPharma Platform

Thermo Fisher Scientific

The Thermo Scientific™ Q Exactive™ Plus and Q Exactive™ HF Hybrid Quadrupole-Orbitrap™ mass spectrometers adds superior denatured and native MS intact analysis and subunit top/middle-down analysis capabilities to the most powerful benchtop peptide mapping instruments available. When combined with Thermo Scientific™ BioPharma Finder™ software it provides a complete integrated hardware and software solution for BioPharma characterization.   The Thermo Scientific™ Q Exactive™ BioPharma platform offers distinct operational modes that have been optimized for the top 3 protein characterization workflows: High Mass Range (HMR) mode was designed specifically for intact therapeutic protein workflows with the aim of providing ease of use an flexibility as needed. HMR mode is optimized for both conventional intact monoclonal antibody analysis performed in denaturing conditions, as well as native MS analysis which is ideal for antibody drug conjugates (ADCs) and other heterogeneous antibody samples Protein mode on the Q Exactive HF is a powerful combination that has set the standard for the highest quality isotopically resolved data achievable on a benchtop mass spectrometer. Deconvolution of subunit MS spectra is easy and powerful with Thermo Scientific™ BioPharma Finder™ Software Peptide mapping with Standard mode is state-of-the-art with the Q Exactive BioPharma, which is a culmination of decades of leadership in the development of data dependent acquisition (DDA)

LC-MSLC-MS (liquid chromatography-mass spectrometry) systems and equipment are used for separation and quantitative analysis of complex mixtures, combining liquid chromatography and mass spectrometry. Quantify proteins, contaminants, pesticides or screen for drug metabolites with a high level of sensitivity. LC-MS systems and equipment include reverse phase, normal phase and specialized columns integrated with various MS detectors such as time-of-flight (TOF), quadrupole, orbitrap or ion trap mass analyzers. LC-MS/MS instruments equipped with a qTOF or triple quadrupole analyzer give greater sensitivity and resolving power to your analysis. The nano refers to the very low flow rate of the mobile phase in the system. The advantage of nanoLC is its high sensitivity. Often the system is integrated with a mass spectrometer in nano LC-MS or nano LC-MS/MS. When using nanoLC equipment consider column choice, software and accurate and precise flow rate control.ProteomicsProteomics is the systemic bioinformatics study of proteins and amino acids, including their structure, size, function and identification. Tools used in proteomics include chromatography, blotting and gels, protein arrays, mass spectrometry and ELISA and associated analysis software. Analyzers and proteomic systems should be sensitive, high resolution, fast and may be automated for high-throughput.Protein BiologyThe analysis of protein expression, identity and function is vital for many areas of life science research and drug discovery. Some of the most commonly used techniques in protein analysis include Western blotting, electrophoresis and mass spectrometry.High ThroughputHigh throughput experiments allow the simultaneous processing of several samples. This parallelization reduces the cost per experiment and increases reproducibility and output volume of data.LC-MSLiquid Chromatography-Mass Spectrometry (LCMS) is a powerful analytical technique that combines the separation power of liquid chromatography with the detection capabilities of mass spectrometry. It is widely used for qualitative and quantitative analysis of complex mixtures in pharmaceuticals, proteomics, and environmental studies.
