Provide quick, robust and convenient application
Peptide map analysis using tryptic digests
The Smart Digest using immobilized trypsin provides a quick and convenient digestion tool for monoclonal antibodies (in 15-30 min). If full coverage is not a goal, no need for additional reduction/alkylation. Resulting peptide mixture can be used for peptide map analysis or glycosylation analysis focusing on glycopeptides. With post-digestion reduction/alkylation full coverage can be achieved.

quick digestion of proteins
Ideal when looking at PTM's in the target proteins. Longer peptide generation protocols, that can be cheaper per sample, will inevitably introduce unwanted PTM's, especially oxidation of methionine that can interfere with downstream analysis. Not ideal for phosphopeptide analysis as the buffers lack phosphatase inhibitors.

Selecting Buffers to Remove Uncertainty in Tryptic Digestion
Protein digestion is a fundamental technique employed in biopharmaceutical and proteomic applications. It is used to analyze the sample and identify structural features or post-translational modifications (PTMs). Despite its widespread use, protein digestion still provides many analytical challenges. This application note demonstrates how Thermo Scientific™ SMART Digest™ kits remove uncertainty associated with conventional solution-based tryptic digestion protocols, resulting in higher reproducibility and sample characterization.
Increased Efficiency of Biomolecule Identification by Optimization of Trypsin Digestion Buffers
Protein insolubility can be a major impediment to traditional bottom-up proteomic methods because it reduces the efficiency of enzymatic digestions and makes analysis problematic. Insolubility can be caused by a variety of factors, including the salt concentration, the presence of various organic solvents, temperature, or simply the nature of the protein itself. This application note demonstrates how the Thermo Scientific™ SMART Digest™ kit removes the uncertainty associated with conventional solution-based tryptic digestion protocols, resulting in higher efficiency and sample characterization.
Improvement in Speed and Reproducibility of Protein Digestion and Peptide Quantitation, Utilizing Novel Sample Preparation Technology in a Full Solution Workflow
Traditional trypsin digest protocols are time consuming, with some protocols taking up to one and a half days to complete and involving multiple steps including evaporation and reconstitution. This adds potential for a high degree of variability. This application note describes a rapid and precise protocol for peptide analysis utilizing immobilized, heat stable trypsin and micro-elution solid-phase extraction (SPE) coupled with next generation ultra-high pressure liquid chromatography and high resolution mass spectrometry (UHPLC-HRMS).
Enhanced Digestion for Improved Biomarker Identification
This application note describes how peptide mapping was used to identify biomarkers of interest, before time-course studies were run to optimize digestion time of thyroglobulin.
Streamline Your Protein Digestion with the Thermo Scientific™ SMART Digest™ Kit
Watch this video to hear Dr Mike Oliver, Sample Preparation Product Manager at Thermo Fisher Scientific, explain how the SMART Digest kit enables greater speed, reproducibility and sensitivity for better sequence coverage over traditional in-solution digestion. The kit uses immobilized trypsin to rapidly digest proteins without sacrificing data quality or reproducibility and takes minutes to run, making it ideal for high-throughput labs, particularly in the biopharmaceutical industry.
