The Oasis cartridges are very easy to use, robust and reproducible. The selection of an appropriate purification protocol increases the specificity leading to a very clean extract.

The Oasis µplate was used as sample preparation for LC TOF application in order to get a valid concentration determination. The product was very easy to use and produced reliable results and good quality.

Solid phase Extraction of biological samples to concentrate the metabolites

The PRIME HLB microElution Plate is a great apparatus for the Solid Phase Extraction technique. I used it to concentrate my metabolites of interest in rat urine samples, and it worked perfectly. But, it didn't work as effectively while concentrating the same metabolites in mice plasma samples. I believe the reproducibility across species (rats vs mice) and the kind of biological samples (plasma vs urine) is low. I would like to see better optimization of the apparatus.

Sample preparation for analysis of THC and metabolites in whole blood from impaired drivers

After evaluating many of the SPE products currently on the market on the criteria of recovery, matrix cleanup, ease of use, and cost, Oasis PRiME HLB uElution plate by far is the best option for the analysis of drugs of abuse in whole blood samples. Waters provided excellent support through application notes, in-person training and method development, troubleshooting, and equipment support throughout the optimization and validation process. The uElution plates demonstrate excellent reproducibility, recovery, and matrix cleanup, even with a complex matrix such as whole blood. The 96-well plate form factor will support a lot of scalability for our lab as we receive additional samples, and the Waters positive pressure manifold makes sample processing extremely easy and rapid. Overall, a great system for tricky analytes and matrices!

Application Area:

Metabolites in urine samples

We are using the Oasis HLB µElution Plate 30µm for our Porphobilinogen analysis in urine and the Oasis MCX µElution plate for our LysoGb3 analysis in urine, on our metabolic lab. We have a manifold voor the extraction system, also from Waters. We use it to concentrate the components to detect them on our Waters Micromass Quattro Micro. We are performing this analysis for a few years now and it gives great results for us. The lysoGb3 analysis is in nmol/l. So the limit of detection is very good for us. We had a private workshop on our lab by some employees from Waters to show us how it all works. So we are satisisfied with it! The plates are not cheap, but they work for us!

A comparison of Extraction Techniques in Bioanalysis and Forensic Toxicology Analyses

In this application note, a comparison was performed between Oasis PRiME HLB SPE, SLE and LLE in both plasma and urine matrices for bioanalysis and forensic toxicology. In plasma, 22 comonly analyzed pharmaceuticals, steroids, and drugs of abuse were extracted using the three aforementioned methods and the results were compared. In urine, 23 drugs of abuse representing opioids, stimulants, benzodiazepines, and synthetic cannabinoid metabolites were tested for forensic toxicology analysis.

This application note demonstrates the need for robust and sensitive analysis of peptide species challenges, both through chromatographic separation and mass spectrometry. Recombinant human insulin and its analogs are perhaps the best known and most widely sold biotherapeutics. Historically, such biologics have been quantified using ligand binding assays. However, specifically in the case of insulin and analogs, these affinity-based assays lack standardization, are subject to matrix effects, and in some cases lack adequate specificity. The use of ionKey/MS facilitated a reduction in sample volume and concomitant increase in sensitivity for the quantification of human insulin and 5 important analogs.

Simultaneous Analysis of Intact Human Insulin and Five Analogs in Human Plasma Using μElution SPE and a CORTECS UPLC Column

Insulin is perhaps one of the best known and earliest peptide therapeutics. Multiple long and fast-acting analogs have also been developed, and a patient may often be prescribed one of each for diabetes control. Quantification of biologics, such as insulins, has historically been performed using ligand binding assays (LBAs) such as ELISAS. LC/MS/MS, however, has certain advantages over LBAs, such as shorter development times, higher accuracy and precision, the ability to multiplex, no cross-reactivity, and the ability to readily distinguish between closely related insulins. This work provides a single, simple method for the simultaneous, direct quantification of intact human insulin and multiple insulin analogs in human plasma using mixed-mode solid-phase extraction (SPE) and a high-efficiency, solid-core particle column that contains a low-level positive surface charge.

Improving a High Sensitivity Assay for the Quantification of Teriparatide in Human Plasma Using the ionKey/MS System

This study combines μElution SPE and the novel and highly efficient ionKey/MS system to improve a quantitative assay for teriparatide in human plasma. Teriparatide is a recombinant form of a fragment of human parathyroid hormone, used in the treatment of osteoporosis. Peptides in general are often difficult to analyze by LC-MS/MS, as MS sensitivity is low due to the formation of multiple precursors and poor or overly extensive fragmentation. IonKey/MS can provide increased sensitivity compared to 2.1 mm I.D. chromatography with the same injection volume, or equivalent or greater sensitivity with reduced sample consumption, making it ideal for peptide analyses.
