Learning the scripts at first was a little complicated but once a script is written and validated, it truly is a walkaway instrument. It transferred over 25000 samples with almost 0 variation on liquid levels, it’s very accurate.

Library production for NGS utilizing NEB chemistries
There is a very steep learning curve, but once you've accomplished that the instrument works well. It took us almost a year to produce our first library. One of the problems (not Beckman's) is that NEB continues to change their protocols and it is very difficult to reprogram volume changes (Beckman's problem) the way the software works. It's similar to using HTML code rather than being able to simply make changes in the software available to the end user. These types of changes have to be done by the Beckman field rep and they are reluctant to pass along that information.

The software needs some work to be more user friendly. Low volumes can be challenging and sometimes tips do not connect properly - so take care on the tip choice and the tip head alignment is critical.

For small to medium-sized laboratories, the Biomek 4000 Laboratory Automation Workstation offers an expanding portfolio of validated and demonstrated NGS sample preparation methods. The methods for PCR setup and AMPure XP cleanup provide researchers with a low cost solution for automating critical steps in the NGS sample preparation process that reduce errors and are proven to generate quality results. For Laboratories with higher throughput needs, automated solutions are available on the Biomek NX P and Biomek FX P Workstations.
There’s a growing library of methods for applications from gDNA library construction to RNA sequencing and capture protocols. Built on flexible Biomek Software, the methods address the most tedious NGS workflow steps including nucleic extraction, library construction, size selection, PCR/qPCR setup, AMPure XP PCR cleanup, normalization and pooling. Beckman Coulter’s library of NGS methods include intuitive Graphical User Interfaces that enable quick implementation so new users can set up and run automated NGS sample preparation with minimal training. The methods are also written to maximize walk-away time, freeing time to work on other tasks. Analyzing the Effect of Pipette Tip Geometries on Fluid Velocity and Shear Strain Rate
Sample preparation protocols specify the need for “wide bore” pipette tips and cannulas to minimize the inadvertent harm, which can be applied to native biological sample matrices during sample preparation. Loss of biological sample integrity can occur due to the pipette tips’ small internal orifice diameter and the speed of aspiration and dispense during pipetting. Biomek Wide Bore Pipette Tips have been designed to address such sample preparation challenges. Through the use of Computational Fluid Dynamics (CFD) software, physical forces and flow characteristics within a pipette tip during pipetting have been simulated. Comparisons of pipette tip orifice diameters can be analyzed for such parameters as fluid (sample) velocity, wall shear, and shear strain rate.
