Excellent system if you are working in a field where gene quantification needs to be measured with high sensitivity in an accurate manner. Great machine, easy to handle and to maintain. I am grateful to have this system in our workplace for the great results.

Testing a variety of samples for ddPCR analysis
The droplet generator helps precise analysis with clear droplet units. When it is working well, it can take a lower limit of DNA concentration for successful testing. It is easy to use since all of the steps of droplet generation are automated after you start your run. However, it occasionally reports error messages and takes multiple running cycles to recover from causing an error.

The AutoDG Instrument partitions each sample in a 96-well PCR plate into ~20,000 uniform nanoliter-sized droplets in which nucleic acid molecules are randomly distributed. Every droplet serves as an individual PCR reaction in Bio-Rad’s Droplet Digital (ddPCR™) PCR System. After partitioning, the plate is sealed with the PX1™ Plate Sealer, and PCR is run to end point in a thermal cycler. The droplets in each sample can then be analyzed using a QX100 or QX200 Droplet Reader.
The AutoDG Instrument eliminates user-to-user variability, allowing you to fully exploit the precision of ddPCR technology by providing more consistent droplet counts across users and plates. The AutoDG Instrument also minimizes hands-on time: it takes less than 5 minutes to load the instrument, and no supervision or user activity is required during droplet generation.

Multiplex Screening of KRAS Mutations in Cell-Free DNA Colorectal Cancer Samples
This application note describes a ddPCR KRAS Screening Multiplex Kit for screening actionable KRAS mutations using DeopletDigital PCR (ddPCR). The ability to screen for multiple KRAS mutations down to 0.2% in a single well reduces the likelihood that mutations will be missed in poor-quality cell-free DNA anf formalin-fixed, paraffin-embedded (FFPE) samples with limited amounts of amplifiable DNA.
