Description

Direct Detection of SAH with a TR-FRET Readout. The AptaFluor®AptaFluor SAH Methyltransferase Assay directly measures SAH produced by methyltransferases during methylation. These enzymes methylate a variety of substrates using S-Adenosyl-L-methionine (SAM) as the methyl group donor, leaving the common product S-adenosylhomocysteine (SAH). AptaFluor overcomes problems with current detection methods that can’t detect enzymes with low turnover or high affinity for SAM. This assay is designed for high throughput screening with an easy-to-use, mix-and-read format. SAH Methyltransferase Assay uses an aptamer, that selectively binds with SAH, the invariant product of methyltransferase reactions. The exquisite affinity and selectivity of the riboswitch combined with a positive TR-FRET signal enable screening and profiling of histone methyltransferases with unparalleled sensitivity.

  • Ultra-Sensitive - The most sensitive HTS methyltransferase assay available. Reduces enzyme usage and allows the use of physiological SAM concentrations as low as 100nM.
  • Stable - Outstanding reagent and signal stability reduces assay variability and allows for automated workflows.
  • Simple & HTS-Ready- The AptaFluor SAH Methyltransferase Assay is in a simple, HTS-ready mix & read format. Run your enzyme reaction, add quench reagent, add AptaFluor reagents, and read your plates.

Interrogating PRMT5, MLL4, METTL3/14, and NSP14 with the AptaFluor SAH Assay

In this application note from BellBrook Labs, explore how the Aptafluor® SAH Assay provides a reliable and robust tool for detecting activity of four methyltransferases (MTs) using various acceptor substrates, including peptides, DNA and RNA. The AptaFluor SAH Methyltransferase Assay is a high-throughput enzymatic assay for SAM-dependent methyltransferases. The assay relies on a naturally occurring SAH-sensing RNA aptamer, or riboswitch, that binds SAH with nanomolar affinity and exquisite selectivity.
