Analysis of Fentanyl and Designer Fentanyl Derivatives in Urine using SPE and HPLC-MS/MS

1 Mar 2018

The United States has seen an increased usage and abuse of fentanyl and fentanyl analogs over the last decade, increasing the list of derivatives that forensic toxicologists need the ability to identify. Meanwhile, typically employed immunoassays have proven to be inaccurate for the initial detection of these novel, synthetic drugs.

In this application note, a universal extraction approach for both current compounds and emerging analytes is illustrated, involving a three-step solid phase extraction procedure for the identification of fentanyl and its major urinary metabolite norfentanyl along with eleven additional synthetic opioid compounds.

Clean Screen® XCEL Solid Phase Extraction Columns

UCT, Inc.

The Clean Screen® XCEL Solid Phase Extraction Columns are a line of sample prep phases designed to reduce the number of steps in the extraction, and therefore the amount of time and solvent necessary to complete the sample cleanup. Further advantages include reduced sample size and improved cleanliness and recovery. The Xcel I column will extract a wide array of basic drugs including benzodiazepines and opiates. The Xcel II column is designed solely for the extraction of the THC metabolite.

UHPLC and HPLCHigh performance liquid chromatography (HPLC) and ultra high performance liquid chromatography (UHPLC), also known as UPLC, are analytical techniques used to separate, identify and quantitate components of complex mixtures including biological samples such as proteins and lipids as well as chemical mixtures of pesticides, drugs and oils. Both techniques are liquid chromatographic methods but differ by operating pressures (HPLC < 6000 psi < UHPLC ). Components of HPLC and UHPLC systems include columns, detectors, pumps, autosamplers and column heaters. Explore a range of UHPLC and HPLC columns for your specific sample needs including reverse phase, normal phase, ion exchange, HILIC, ion exclusion and size exclusion columns. For more specialized HPLC, explore FPLC, countercurrent LC and simulated moving bed systems. Multiple formats are available for conducting SPE, including prepacked SPE cartridges, disks and microplates, as well as SPE sorbent powders for manual packing. SPE systems can be used to automate the process and extract multiple samples at once. Solid-phase microextraction (SPME) and supported liquid extraction (SLE) products are also available in the same formats. Techniques used include HPLC, chromatography, spectroscopy, mass spectrometry, immunochemical, electrophoresis, turbidometric / spectrophotometric assay, MRI and ISE analysis. Tests are often carried out on plasma or serum but urine (urinalysis) and fecal specimens are also processed.Evidence Collection and AnalysisEvidence Collection and Analysis is the retrieval and subsequent investigation into criminal evidence obtained from a crime scene. Forensic genetic instruments, kits and reagents enable the analysis of evidence collected  at the crime scene. Reagents and kits for presumptive analysis for body fluids such as blood, saliva and semen in addition to drugs and narcotics test kits are used for CSI. Fingerprinting equipment, forensic light sources, casting kits and equipment for collecting recoverable materials can be found in the directory.Toxicology / Drugs of Abuse TestingToxicology and Drugs of Abuse Testing is the investigation into toxic and illegal substances found at the scene of a crime or from a suspect and / or victim associated with a scene of a crime. Analyzers, immunoassays and drugs of abuse test kits and presumptive test kits are an essential aspect of toxicology investigation.Drugs of AbuseA drug of abuse is a drug that has been taken for non-medical purposes. Such drugs can have mind-altering effects, with some causing dependence and addiction.Forensic InvestigationForensic investigation is a diverse collection of scientific techniques and methods for the identification and characterization of chemicals relating to crime scene investigations.Forensic ToxicologyForensic toxicology is used to determine cause of death, drug use and contaminations. Toxicological evaluations are commonly used in analytical chemistry, clinical, life sciences and forensics.
