Rapid Determination of DNPH Derivatized Carbonyl Compounds

This application note by Knauer presents a fast method for the determination of six carbonyls. Low molecular weight aldehydes are volatile and polar substances, hindering analysis by RP-UHPLC methods. Also, many carbonyl compounds lack chromophores and cannot be detected by a UV detector. However, functional carbonyl groups allow aldehydes and ketones can be derivatized. Derivatzation with DNPH forms a coloured mixture of DNPH-carbonyls with a lower vapour pressure which can be separated by UHPLC and detected with a UV source.
