Assay of ASIC1a Channel with the IonWorks Barracuda System

In this application note, an assay is presented of acid-sensing ion channels (ASICs) using the IonWorks Barracuda Automated Patch Clamp System. Electrophysiological recordings were made from Chinese hamster ovary (CHO) cells stably transfected with human ASIC1a ion channel. The ASIC1a currents were elicited by rapidly replacing the regular external buffer with acidic buffer. Concentration-dependent inhibition of ASIC1a currents by amiloride and benzamil were also measured.
