  • Application Area: Developing SNP markers in tilapia species using double digest RAD sequencing

"T4 DNA ligase is very effective enzyme to joining DNA fragment produced from cutting site of enzyme restriction SbfI and SphI to a barcode sequences. The barcode can be perfectly recalled in the analysis after sequencing the DNA library."

Thermo Scientific T4 DNA Ligase is a single polypeptide. It is commonly used to join DNA molecules to each other. T4 DNA ligase catalyzes the formation of a phosphodiester linkage between DNA chains by condensing the 5'-phosphoryl group of one DNA chain with the adjacent 3'-hydroxyl group of the other DNA chain. T4 DNA ligase will ligate blunt and homologous cohesive ends of double-stranded DNA. It is the only DNA ligase that will efficiently ligate blunt ends in the absence of macromolecular exclusion molecules. T4 DNA Ligase can repair nicks in duplex DNA, RNA or DNA/RNA hybrids. It can also join RNA single strands via bridging oligonucleotide adaptors.
