Offers a number of partitioned reactions but not compatible with qPCR applications
Rating: 5.0

  • Application Area: validation of microarray, copy no., SNP

"The larger number of reactions (20,000 per sample/1.9 million per plate) generated with this platform increase the precision in determining absolute quantification and CNV measurements. The increased number of reactions per sample affords the ability to load larger amounts of template DNA when compared to the other systems that have fewer partitioned reactions per sample. This becomes increasingly valuable when detecting rare events of importance. Digital reads from duplicated wells can also be combined to determine CNVs for rare or low copy mutational events. Lower limits of detection have been reported to allow identification of 0.001% of the mutant population in partitioned reactions. Bio-Rad's QX100 Droplet Digital PCR platform has also been used with maternal plasma DNA to determine absolute quantification measurements of maternal and fetal markers, underscoring the advantages of dPCR technology in measuring CNVs with low quality/low quantity template DNA. Though the QX100 Droplet Digital PCR instrument offers a substantial number of partitioned reactions per plate (1.9 million) and is reasonably priced."

Review date: 22 Oct 2015 | QX100™ Droplet Digital™ PCR System (186-3001)
The QX100 Droplet Digital PCR (ddPCR) system provides an absolute measure of target DNA molecules with unrivaled performance in precision, accuracy, and sensitivity for quantitative PCR applications.

The Droplet Digital PCR system is the third generation of PCR technology and provides a revolutionary approach to target DNA quantification.

The QX100 system consists of two components: the QX100 droplet generator and the QX100 droplet reader. A Bio-Rad thermal cycler equipped with a 96–deep well reaction module is also needed to complete the Droplet Digital PCR workflow.

The QX100 droplet generator partitions samples into 20,000 nanoliter-sized droplets. After PCR on a thermal cycler, droplets from every sample are streamed in single file on the QX100 droplet reader. The PCR-positive and PCR-negative droplets are counted to provide absolute quantification of target DNA in digital form.

QX100™ Droplet Digital™ PCR System Benefits:

  • Detect rare DNA target copies with unmatched sensitivity
  • Determine copy number variation with unrivaled accuracy
  • Measure gene expression levels with the highest precision

QX100™ Droplet Digital™ PCR System Applications:

  • Copy number variation
  • Rare sequence detection
  • Mutation detection
  • Gene expression analysis
  • miRNA analysis
  • Next generation sequencing sample quantification
