The DNA IQ™ System is a DNA isolation system designed specifically for forensic and paternity laboratories. This system employs novel paramagnetic particles to isolate clean DNA for use with short tandem repeat (STR) analysis. The DNA IQ™ System can be used to extract DNA from a variety of sample types, including stains and liquid samples. Protocols for database samples and casework samples are available.
The unique DNA IQ™ Resin removes PCR inhibitors and contaminants frequently encountered in casework samples. When working with larger sample volumes, such as those found in paternity and databasing, the DNA IQ™ System can deliver a consistent amount of total DNA. Samples including buccal swabs, liquid blood and stains on FTA® and other blood cards have been used with the DNA IQ™ System. Some samples, such as tissue and hair, require pretreatment with proteinase K. In addition, extracting DNA can be difficult from substrates such as denim, envelopes, carpet, etc. The Tissue and Hair Extraction Kit increases DNA yield from challenging casework samples and substrates. The kit uses Proteinase K and other reagents to remove proteins and other components from the DNA, helping to ensure maximal yield and recovery of DNA from casework samples. The DNA then can be purified using the DNA IQ™ System.
The DNA IQ™ System has been tested with the Plexor® HY System and PowerPlex® Systems to ensure a streamlined process. This translates into reliable products that give optimal results from isolation to quantitation and STR analysis.
Genomic DNA isolation using the DNA IQ™ System has been automated on the Biomek® 2000 and 3000 laboratory automation workstations, as well as the Tecan Freedom EVO® liquid handler. Please contact Promega Technical Services for additional information.

Comparing Five Forensic DNA Extraction Methods
Many different extraction protocols are used in the forensic community. Typically these extraction methods work well for the majority of samples encountered. However, not all extraction methods perform equally well with heavily inhibited samples. This scientific poster evaluates the efficiency of several commonly used protocols. Each extraction method had to have the ability of organic extractions to reduce the amount of inhibitors being co-extracted, while still obtaining a suitable quantity of DNA for STR analysis. To evaluate the efficiency of the procedures, both quantity and quality of DNA were compared. The ability to automate each of the methods is also discussed.
High-Throughput Genetic Data Gathering from Bone Samples Recovered from an Air Crash
DNA-based analysis constitutes the key approach in the challenging task of identifying human remains from mass fatalities. In this application note a workflow that allowed effective and fast identification of the complete set of highly fragmented human remains (N = 418) from an air crash in Río Negro Province, Argentina, on May 18, 2011 is presented. This process included DNA extraction from bone using a semi-automated DNA purification system, DNA quantitation by real-time PCR using commercial kits and data analysis using expert software.
Improved Performance for Forensic Casework: Extraction and Isolation Updates for the Maxwell® 16 Instrument
This application note describes recovery of DNA from casework samples using the Promega DNA IQ™ System on the Maxwell® 16 Forensic System. This method optimizes the efficiency of sample recovery from a substrate and the subsequent isolation of DNA to enable increased DNA yield across a variety of samples.
