Electron Transfer Dissociation of N-linked Glycopeptides from a Recombinant mAb Using SYNAPT G2-S HDMS

Glycosylation analysis is important during the production of recombinant therapeutic antibodies, typically involving a released glycan assay, glycosylation site determination, and site heterogeny analysis. In this application note it is shown that electron transfer dissociation (ETD) is an indispensible tool for characterizing the post translational modifications of biotherapeutic proteins. In addition, data from this application demonstrate glycopeptides site heterogeneity from a monoclonal antibody, trastuzumab, can be achieved using ETD on a SYNAPT G2-S HDMA with post-column mixing of m-NBA.
