Best extracellular matrix to culture intestinal organoids.

"Enteroids growth in a consistent manner and keep the same morphology over the passages. When compared with other ECM from animal origin, enteroid growth in matrigel is more robust and reproducible. Use of matrigel LOTs with low level of endotoxins is preferable. So sometimes you need to wait some time to get a good LOT. Use of matrigel requires some attention since it needs to be manipulated always on ice to avoid early polymerization."

Perfect matrigel for intestinal organoids!

"We have been using this matrigel for intestinal organoids culture and they have been working really well. The viscosity is suitable to form a dome in the wells and make a perfect matrigel pad for intestinal organoids. It is totally transparent and solidifies pretty easily when put in the incubator for 5 minutes! I definitely recommend this for organoid culture!"

Corning® Matrigel® Growth Factor Reduced (GFR) Basement Membrane Matrix, 10 mL, is suited for applications requiring a highly defined basement membrane preparation. Available in standard GFR, High Concentration (HC), and Phenol Red-free formats. Corning Matrigel matrix is a solubilized basement membrane preparation extracted from the Engelbreth-Holm-Swarm (EHS) mouse sarcoma, a tumor rich in extracellular matrix (ECM) proteins, including Laminin (a major component), Collagen IV, heparan sulfate proteoglycans, entactin/nidogen, and a number of growth factors.

Quality

  • Mouse colonies routinely screened for pathogens via Mouse Antibody Production (MAP) testing
  • Extensive PCR testing performed to screen for a number of pathogens
  • Tested and found negative for bacteria, fungi, and mycoplasma
  • Protein concentrations determined by Lowry method
  • Endotoxin units measured by Limulus Amoebocyte Lysate assay
  • Gel stability tested for a period of 14 days at 37°C
  • Biological activity determined for each lot using neurite outgrowth assay
