"The new generation system is very intuitive to use and gives very reproducible results. It is very easy to set up and you are able to make changes to a run quickly on the fly. Switching to reverse phase is also very straightforward and would be useful to know for new potential customers who often work with very polar compounds. After sales care is very good. All problems have been sorted out very quickly through engineer visits."

"Time saver versus packing my own columns. Uses less solvent than regular flash column chromatography and generates less hazardous waste. Since the columns are packed tighter, tough separations are much easier. The ISCO doubles my production of purified compounds. The NextGen can withstand higher pressures which is helpful if my compound crashes out on the column."

Solved a lot of problems even with purifying compounds that have difficult separation!

"It is straight forward and easy to use. Saved a lot of time compared to manual columns. I always forget about my column so it always either dries out at the top or overflows when I'm not looking. Works really well on difficult separation and on TLC as well. I would still suggest taking TLC before hand instead of going in blind. Speed of solvent is also adjustable but I have always used suggested option when you pick your column size. One of the best feature is the real-time monitoring through UV with different wavelength (visible range optional but we did not have that). It automatically advances tubes depending on amount but also when it detects a drop in intensity and increase (like a new compound is coming out). It can also be manually advanced too if you like. Also has 3 options where the collection goes. Can either collect everything in test tubes, none where everything will go into the waste bottle or peaks where they only collect when they detect the peaks. At anytime of the run, you can change the gradient easily from the screen. It also has sensor to monitor pressure of the system and the solvent reservoir. Just a tip, make sure your compound is fully soluble if you're doing liquid loading. Otherwise it will get stuck and the pressure will overload. Overall, I really like it compared to manual columns where easy separation can take as little as 20 mins and my compound is pure in NMR."

"The Teledyne ISCO CombiFlash NextGen300+ is a workhorse in our synthetic lab. It can be used to purify from a milligram to about 100 grams of sample. It gets used on nearly a daily basis, often multiple runs in one day and it holds up well. It is easy to clean and prep for a new run, although a programmable cleaning protocol would be nice. The instrument works well and holds up to a lot of different uses (normal and reverse phase, specialty columns, hand-packed columns, solid and liquid loading, etc.). The unit has a fairly intuitive interface, and if you have used a Teledyne purification system then operating this one will be very easy. After sales care is pretty good. When I have needed to make repairs, help from the technical support line is quick and professional. I have had mixed luck with help diagnosing problems, but parts are delivered quickly when needed. My chief complaint with the system is that the accessories are very expensive. Getting different size column cartridge caps, extra racks, and external column adapters is very pricey. Regardless, the NextGen system itself is a good value for the money. You won't find a better unit at the price point."

The system is fitted with a solid load injection valve that can be bypassed if you decide to perform a liquid injection. RFID technology comes standard and automatically reads the size and type of RediSep or RediSep Gold column being used for the purification. Based on this information, a default gradient and flow rate are programmed into the system. Simply insert the column into the stand, press Play and the run starts. This same RFID technology automatically reads the rack type being used ensuring there is never a missed or overfilled tube. Active solvent and waste level monitoring is standard on the NextGen 300+.
