Application Note: Multidimensional LC-MS-Based Lipidomics Using the SCIEX TripleTOF® 5600 LC-MS System

This application note presents the concept of Multidimensional Liquid Chromatography MS- (MDLCMS-)based lipidomics applied to lipid extracts from liver and skeletal muscle of mice fed a normal or a high fat diet, according to the Multidimensional Mass Spectrometry- (MDMS-)based shotgun lipidomics approach. MS analyses of lipid extracts massively suffer from ion suppression effects limiting the detection of low abundant lipid species. Exemplarily, the following 8 dimensions for lipid detection and identification were used: chromatographic separation using 3 different conditions, acquisition of each in positive and negative ionization mode, high resolution MS and MS/MS. The combination of high resolution, mass accuracy, sensitivity and speed of MS and MS/MS scans using the AB SCIEX TripleTOF® 5600 LC-MS System enabled rapid comprehensive analysis of a lipid extract in only 1 hour.
