"xCELLigence is incredible. So easy to use. Very user friendly. It also can be used for almost ANY cell type: unlike other systems it does not require intact monolayers. Also, something like 70% of the well is electrode, making it incredibly sensitive to even small fluctuations."

"This is one of the best and highly sensitive instrument for cell proliferation assays. We use this instrument a lot. The ease of setting up the assay and watch the resulting graphs without having to do anything for days together is simply awesome. Unlike end-point assays this continuous monitoring of cell growth/death gives a better understanding of the biological process. The highlight of this instrument is its small and does not take up the whole incubator space. The software used is also very user friendly and setting up the assay does not take more than couple of minutes. The only downside of this instrument is the cost of the plates used."

Great device, annoying software

"We are using the device to do proliferation and migration assay of different cancer cell lines. The device is easy to handle. Although, I don't like the software of the Xcelligence. Espacially that you can't safe the data analysis part and also that you always have to click newly what you want to see on the screen, when you open a new file. Most of our results are reproducable. Sometimes I find it difficult to interpret the results. Therefore, I would find it cool when there would be some kind of forum where users could discuss their problems and exchange tipps and tricks. Otherwise the support from the OLS in Basel is very good, they always answer our questions fast and detailed."

"Lable-free and real-time analysis of cell growth and behavior made simple with xCELLIgence RTCA DP. Good for short term response as well as long-term behaviour. Can accomodate various cell lines which showed consistent results. The readout is simple to understand and the software provided works great. Although the E-plate is expensive, you can get solid results, so value for money is not bad. I got after sales care for the first two years, which would be enough because the machine is not complicated and does not need much care except regular cleaning and common sense maintenance."

"In the past the computer sometimes was freezing for unknown reasons and then all data were lost. However, recently everything runs smoothly. Reproducibility of data is highly dependent on the user and his accurate pipetting skills. Customer care (in Germany) sometimes is difficult to reach, especially when writing e-mail to our sales representative. However, if reached, support is very competent and helpful."

The fact that you can monitor killing over multiple days makes it a must-have

"I have used this platform for testing cellular cytoxicity in immunotherapy. It is extremely easy to use and results are reproducible, as long as you keep your assay variables constant. Otherwise the high sensitivity of the platform will play tricks on you. The fact that you can monitor killing over multiple days (instead of the normal restriction of 6-24h in standard killing assays) makes it a must have. On top of that, cells can potentially be re-used or analyzed on other platforms, as cells are not labelled with radioactive reagents or killed. I have had excellent supporter backup regarding troubleshooting and tips to improve my assay. All in all, this platform is highly recommendable for studying cellular toxicity."

"The xCELLigence DP is easy to use, provides the flexibility of monitoring cells in real-time and allows the assessment of multiple time points and dual functions in a single experiment, without the need to repeat tedious, conventional endpoint assays numerous times. The small reagent volumes required supports the economical use of limited reagents. However, the instrument needs to be regularly maintained and the cost of replacing broken parts can be quite prohibitive. Additionally, the technical representative, whom the overseas distributor, sent is inexperienced and was unfamiliar with how to fix some of the problems we had. Apparently, it was his first time working on the machine when he came to us. "

"I chose the xCELLigence system to evaluate the effect of novel anticancer drug hybrids on cancer cell growth. Overall the system was very user friendly and graphs generated were intuitive, informative and reproducible. This platform offered several advantages specific to my research needs. Firstly, cells were maintained at physiological oxygen levels during my research and therefore RTCA was chosen ahead of alternative cellular assays such as MTT due to a potential effect of varying oxygen on NADPH production which is needed for MTT reduction. Furthermore, RTCA detected not only drug-induced changes in cell number but also the kinetics of drug action. RTCA was invaluable in defining optimal seeding densities and drug concentration range and enabled us to detect instances of cell recovery which may otherwise have been overlooked. The IC50 function was also useful in surmising overall activity of novel anticancer drugs. "

"Our lab is using the xCELLigence DP device pretty often. It could help us to solve various questions for different types of cells. Some of this questions could not be answered by common end-Point assays, why we made some new findings. Once we had the problem, that the USB port of our device started to get rusty although we were using the device normally in our incubator. From our point of view, something like this should not happen, because the xCELLigence device is made for using it under normal conditions, which includes the CO2 incubator. Therefore we called the Service. They made us an offer for about 200 € to exchange the USB-port. We rejected this offer, because we felt that this USB-port is an error made by the Company and not our mistake (with normal use). Expect from the problem with the USB-port, we would recommend to everybody interested in cell behaviour to buy this xCELLigence device. And we are advertising the use of it, to other persons. "

The Agilent xCELLigence Real-Time Cell Analysis (RTCA) DP (dual purpose) instrument continuously measures cell invasion and migration (CIM) and monitors cell health, behavior, and function using label-free cellular impedance.

The three cradles of the DP instrument enable three separate electronic 16-well plates to be controlled and monitored in parallel or independently of one another. This flexible plate batch processing allows maximum productivity for multiple users. The instrument operates in a standard CO2 cell culture incubator and the control unit is housed outside the incubator.

Online data acquisition and offline data analysis are easy to perform using RTCA Software Pro, which also supports FDA 21 CFR Part 11 compliance.
