pV2- Rluc(h)/pGFP2-â-arrestin2 Vector Kit
The V2 Vasopressin/â-arrestin2 vector kit can be used to perform G protein coupled receptor (GPCR) functional assays using BRET2 assay technology. The assay is based on the detection of the agonist-dependent interaction of the GFP2-â-arrestin2 fusion protein with the C-terminus of a GPCR-Rluc(h) fusion protein where Rluc(h) is codon humanized Renilla luciferase and GFP2 is a mutant of the Green Fluorescent Protein. The vector kit includes (1) an expression plasmid encoding a V2 Vasopressin Receptor-Rluc(h) fusion protein and (2) an expression plasmid encoding a GFP2-â-arrestin2 fusion protein.
pV2 Vasopressin Receptor-Rluc(h) Fusion Vector
The pV2 Vasopressin Receptor-Rluc(h) Fusion vector, which expresses a V2 Vasopressin Receptor-Rluc(h) fusion protein, has been designed to serve as the Donor moiety in a BRET2 V2 Vasopressin/â-arrestin assay. In this vector, the Rluc(h) is fused to the C-terminus of the pV2 Vasopressin Receptor (GPCR). The fusion gene is placed under the control of the CMV promoter thus assuring a high level of expression in mammalian cells. When HEK-293T cells co-transfected with V2 Vasopressin Receptor-Rluc(h) and GFP2-â-arrestin2 (cat. # 6310176) are stimulated with an agonist such as [Arg8]-Vasopressin (8-AVP), dose-dependent BRET2 signals are obtained in the presence of the Rluc coelenterazine substrate, DeepBlueC.
