Specificity

The vial contains a mixture of 1.7 – 2.2 µm Yellow Fluorescent Particles that are dyed to have an excitation maximum of approximately 470 nm with a subsequent emission maximum of approximately 480 nm. This particle mixture is useful with flow cytometers equipped with a 488-nm argon laser. This particle mixture has compatible excitation and emission spectra with the fluorescein isothiocyanate (FITC) fluorophore.

Preparation and Storage

The particle mixture (~ 1.0% w/v) should be stored undiluted at 4°C and protected from direct exposure to light. Do not freeze.

Usage

This particle mixture is useful for routine calibration of flow cytometers. Before use, resuspend the particles by vortexing. Dilution of 3 - 5 drops of particles to 1 ml of sheath fluid will provide an adequate number of particles for flow cytometric analysis. Since applications vary, each investigator must determine dilutions appropriate for individual use.
