SMART-Seq v4 Ultra Low Input RNA Kit for Sequencing—Single-Cell Transcriptome Analysis with Ultimate Sensitivity
The SMART-Seq v4 Ultra Low Input RNA Kit for Sequencing is the fourth-generation of SMARTer Ultra Low kits, and our most sensitive mRNA-seq kit for single cells and ultra-low inputs. This kit takes SMART (Switching Mechanism at 5’ End of RNA Template) technology to the next level by incorporating and improving upon the SMART-Seq2 method and locked nucleic acid (LNA) technology. With these additions—referred to as ""SMARTer-Seq® chemistry""—this kit is able to outperform both the SMART-seq2 method and previous generations of SMARTer kits by identifying the greatest number of transcripts. The Clontech tradition of continuous innovation extends to this kit by providing direct cDNA synthesis from as few as 1–1,000 intact cells (or as little as 10 pg–10 ng of total RNA), high reproducibility, even gene body coverage, and excellent representation of GC-rich transcripts. The full-length cDNA libraries from this kit are compatible with both Illumina® and Ion Torrent sequencing platforms.
The core SMART technology in this kit is an immensely powerful tool for studying the full transcriptome, including splice junctions and alternative splicing events in cells, and provides full-length gene coverage. Oligo(dT) priming is used to specifically amplify mRNA from either high-quality total RNA (RIN >8) or intact cells. LNA technology improves the efficiency of template switching, which increases the number of genes identified to higher levels than any other SMARTer Ultra Low kit for mRNA-seq.
