Fast and precise information on formulation of therapeutic antibodies.

"Our company 2bind uses the nanoDSF to optimize the formulation of therapeutic antibodies. The Prometheus allows us to screen 48 buffer conditions with a bit more than an hour. The antibody can be used as it is - without any modification - and in a very broad concentration range. Due to the capillary format the method consumes only low sample material. NanoDSF allows us to study thermal stability (Tm) and aggregation onset temperature within one experiment. The high data point density enables to see even smallest transition points. Output data are highly comparable to DSC, but obtained faster, with less material and less costs. The Prometheus device is very robust and occupies only small lab space. We can definitely recommend this technology as nanoDSF has become a standard application in our lab."

Prometheus NT.48 is based on nanoDSF, an advanced Differential Scanning Fluorimetry technology, using intrinsic tryptophan and tyrosine fluorescence to measure protein stability with ultra-high resolution.

Thermal or chemical stability can be investigated in a label-free setup by detecting changes in tryptophan or tyrosine fluorescence. NanoTemper's dual-UV detector monitors emission at 350nm/330nm allowing on-the-fly fluorescence detection, and provides unmatched scanning speed and data point density. Consequently, the resulting unfolding curves display ultra-high resolution. nanoDSF allows the detection of multiple and minute unfolding signals, since the measurement is not affected by aggregation. As no secondary reporter fluorophores are required, protein solutions can be analyzed independent of buffer compositions, with a high dynamic protein concentration range from more than 250mg/ml down to 5μg/ml. This enables the analysis of detergent-solubilised membrane proteins and highly concentrated and viscous antibody formulations.
