Vectors for High-throughput Cellular Protein Trafficking Assays
ProLabel™ Expression and Cloning Vectors offer convenient and flexible options for developing homogeneous, single-addition PathHunter™ screening assays to directly measure the effect of test compounds on protein translocation, secretion, or degradation by ß-galactosidase activity in whole mammalian cells. ProLabel Vectors can also be used to monitor general protein expression in a convenient antibody-free format.

All ProLabel Vectors are designed with the 4-kDA ProLabel tag and the CMV promotor for constitutive protein expression in mammalian PathHunter EA-Nuc or in-house Cell Lines. If transfecting a ProLabel Vector into an EA-Nuc Cell Line for a PathHunter Assay, PathHunter Detection Reagents are purchased separately. For general protein expression assays, in-house cell lines may be used and EA must be added using EFC Detection Kits.

For membrane-based detection, expressed ProLabel-tagged proteins can be monitored using the fast, antibody-free EAstern Blot Assay Kit, or if ProLabel-c-myc vectors are used, by traditional Western blots.

Wide Selection of Vectors
ProLabel Expression Vectors are available with the ProLabel tag on a wide variety of proteins known to translocate to the nucleus. In addition, ProLabel Cloning Vectors are engineered for cloning a selected gene fused to the ProLabel tag into C1-3 or N1-3 frames.

PathHunter Assay Principle
When a ProLabel Vector is transfected into cells that express a large fragment of ß-gal in the nucleus (PathHunter EA-Nuc cells), translocation of the fusion protein to the nucleus enables two ß-gal framents to combine and generate signal when chemiluminescent substrate is present. In general non-PathHunter EFC assays, the EA protein is added as a reagent instead of nuclear expression.
