The pGFP2-Rluc(h) vector allows for the expression of a fusion protein between GFP2 and Renilla Luciferase when transfected in mammalian cells. Once expressed, the fusion protein serves as a BRET2 positive control. This vector shows two major differences as compared to pBRET+, the original positive control vector. First, it encodes the codon humanized version of the luciferase gene allowing for higher level of expression in mammalian cells. Also, the presence of GFP2 at the N-terminal portion of the luciferase results in an improved energy transfer efficiency upon addition of DeepBlueC. GFP2-Rluc(h) is placed under the control of the cytomegalovirus (CMV) promoter thus assuring a very high constitutive expression in a variety of cells.
