"I have used this enzyme for almost all kinds of PCR reactions, in particular in site-directed mutagenesis. The enzyme is very stable and can easily amplify long templates - I have used it for amplifying ~ 10 kb plasmid with GC rich sequence. It is also faster than most other DNA polymerases having a speed of nearly 1kb/30 sec. In addition, the enzyme also comes with optimized buffer. However on the downside, it is expensive."
The new Affibody®-based inactivation method of Phusion DNA Polymerase increases the specificity of PCR amplification by preventing non-specific extension of DNA substrate at ambient temperatures prior to the first cycle of PCR.
In addition to the extreme fidelity, speed and robustness, Phusion Hot Start DNA Polymerase now also delivers extreme specificity to PCR experiments. Phusion Hot Start DNA Polymerase does not require a separate activation step in the PCR protocol.
