Cell based kinases using PhosphoQuest PhosphoQuest Assays use monoclonal antibodies which are immobilized on a microtiter plate to bind the phosphorylated specific protein as a standard or a sample. After a short incubation the excess sample or standard is washed out and a rabbit polyclonal antibody to phosphophorylated specific protein is added. This antibody binds to the phosphorylated specific protein captured on the plate. After a short incubation the excess antibody is washed out and goat anti-rabbit IgG conjugated to Horseradish peroxidase is added, which binds to the polyclonal phospho-antibody. Excess conjugate is washed out and substrate is added. After a short incubation, the enzyme reaction is stopped and the color generated is read at 450 nm. The measured optical density is directly proportional to the concentration of phospho-protein in either standards or samples.
