Pfu DNA ligase is a recombinant enzyme derived from pyrococcus furiosus hyperthermophilic marine archaebacterium. The recombinant DNA Ligase has a higher ligation specificity and lower background than Tth DNA ligase.

Pfu DNA ligase catalyzes linkages of adjacent 5'-phosphate and 3'-hydroxy ends of double-stranded DNA at 45°C to 80°C, with a temperature optimum near 70°C for nick-sealing reactions. The ligase is highly thermostable and it increases the reliability of LCR by permitting higher melting temperatures.

Features:

  • Featuring higher ligation specificity and lower background than Tth DNA ligase
  • Eliminates LCR template-denaturation step
  • The ligase is highly thermostable and features a half-life of over 60 minutes at 95°C
  • Increases reliability of LCR by permitting higher melt temperatures
  • Eliminates the LCR template-predenaturation step meaning that less enzyme is required for LCR
  • Operates at an optimum temperature near 70°C for nick-sealing reactions
