BD Gentest™ P450 High Throughput Inhibitor Screening Kits are designed to rapidly screen for potential inhibitors of cytochrome P450 catalytic activity. Each kit provides all the necessary components and instructions for measuring the IC50 value for up to 38 test compounds in 96-well plates. These kits can accommodate other plate formats, e.g. 384- and 1536-well, with proper scaling.
Kit Components:
• CYP2D6 + P450 Reductase
• Control Insect Cell Membrane Protein
• Buffer Solution: 0.5 M Potassium Phosphate (pH 7.4), filter sterilized.
• Stop Solution: 0.5 M Tris Base, filter sterilized.
• Cofactors: 1.3 mM NADP+, 66 mM MgCl2, and 66 mM Glucose 6-Phosphate.
• Glucose 6-Phosphate Dehydrogenase: 40 Units/mL in 5 mM Sodium Citrate Buffer (pH 7.5).
• AMMC (3-[2-(N,N-diethyl-N-methylamino)ethyl]-7-methoxy-4-methylcoumarin): Fluorescence substrate, 0.302 mg.
• Quinidine: CYP2D6 selective positive control inhibitor, 0.0006 mg
• AHMC (3-[2-(N,N-diethylamino)ethyl]-7-hydroxy-4-methylcoumarin hydrochloride): Metabolite standard (10 uM in water)
Note:
• Freeze thaw stability: This CYP2D6 inhibitor screening kit was subjected to 10 freeze thaw cycles without a change in IC50 value for quinidine or a significant loss of signal to noise.
• *QC assay (IC50 determination for quinidine) was performed using all the components contained in the current lot kit.
