LiCor Blocking buffer. Greatly enhanced my Western detection abilities.

"I recently found that using the LiCor Odyssey Blocking Buffer (PBS) with 0.1% Tween in both my primary and secondary antibody incubation steps helps to both reduce background and to enhance the primary target. This finding is in comparison with performing both incubations in PBS Tween alone. The added benefit is that following the initial blocking in the LiCor Blocking Buffer, one can advance to the primary incubation directly- skipping over the PBS Tween washing steps that take 15-30 minutes. Results likely vary with primary and secondary antibodies but this helped me greatly using a Roche AntiGFP primary antibody and the IR800 secondary antibody from LiCor."

"The LI-COR blocking buffer is very easy to use for highly reproducible results. Whenever we have question or need help, the service group are always very help. In terms of value for money, it is not that cheap, however it is worth every penny after we compare all the cheaper alternatives. "

"I use Odyssey Blocking Buffer exclusively for all of my western blots. It is much better than using milk for blocking. I never have any problems with background issues and the blots results are clear. It is extremely easy to use and I use it full strength for the blocking step and also for antibody dilutions."

"Odyssey® Blocking Buffers is really simple and easy to use. It can really block nitrocellulose or PVDF membranes against non-specific binding. However, I only used it for Odyssey scanner system. This product can be reused several times, but you have to take care keeping it at 4ºC. This buffer is really good but the price is not so good when compared with dry milk, for instance, when the role of both is basically the same. The fact you can reuse it and sometimes some antibodies are not so good and you really need a higher quality product for blocking, this buffer is recommended."
Odyssey Blocking Buffers are the gold standard for quantitative Western blot detection. They contain no mammalian proteins, for the lowest cross-reactivity with mammalian antibodies and are available in a ready-to-use formulation in TBS (Tris-buffered saline). The TBS-based buffer is ideal for the detection of phosphorylated targets and is suitable for the widest range of targets for Western blot detection. Be sure to optimize your experimental conditions for the best Western blot and assay results. Blocker choice is important for immunoassay success. Blocking buffers enhance sensitivity by reducing background interference, increasing signal-to-noise ratio, and promoting specific binding of the primary antibody while minimizing non-specific interactions.
